Background: During mouse embryonic development, neural stem cells (NSCs) arise at
embryonic day 7 in the neural ectoderm area. They continue to expand in the neural plate,
neural tube, and fetal brain and persist throughout life in several parts of the adult brain. Current
culture conditions developed for the maintenance of NSCs are not optimal.
Hypothesis: 1. NSCs isolated from different developmental stages of mouse embryos are
characteristically different and might use different signaling pathways for self-renewal; 2. There
must be culture conditions that can support long-term maintenance of NSCs isolated from any
developmental stage of the embryo or adult.
Based on the Background and Hypotheses, please design experiments to: 1. identify new
signaling pathways or genes that are involved in NSC self-renewal; and 2. Develop new culture
conditions for NSC maintenance.
The proposal is expected to be exploratory and the use of any radically different approaches is
a plus.
